以白凤菜为试材,采用组织培养方法,研究了氯化汞浸泡时间及不同激素配比对白凤菜茎段灭菌及诱导的影响。结果表明:外植体最适宜的灭菌条件为0.1%的氯化汞溶液浸泡11min,此时污染率最低为41%,外植体成活率为68%,为较适宜的灭菌时间。MS+6-BA3.0mg/L+NAA 0.02mg/L为比较适宜的白凤菜茎段诱导培养基,诱导率达98%,每块外植体分化的不定芽数目达到8.0个;白凤菜的生根培养基为1/2MS+NAA 0.20mg/L。
Using Bai-Feng-Cai as test material, tissue culture method was used to study the effects of soaking time and different hormone ratio on the sterilizing and inducing of stems of Bai-Feng-Cai. The results showed that the optimal sterilization conditions for explants were 0.1% mercuric chloride solution soaked for 11 min, the lowest contamination rate was 41% and the survival rate of explants was 68%, which was the suitable sterilization time. MS + 6-BA3.0mg / L + NAA0.02mg / L was the suitable medium for the induction of stems of Pakchoi with the induction rate of 98% and the number of adventitious buds per explant reaching 8.0. The rooting medium for vegetables was 1 / 2MS + NAA 0.20 mg / L.
