目的对吴茱萸中多糖进行提取、分离和纯化,进一步研究吴茱萸多糖主要组分的单糖组成。方法采用水提醇沉法提取粗多糖,H2O2脱色、Sevag法除蛋白、透析制备精制品,用DEAE-32纤维素阴离子交换柱、Sephacryl S-400 SF型凝胶柱分离纯化多糖,经酸水解后,1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生化HPLC法分析单糖组成。结果从吴茱萸中分离纯化得两个主要多糖组分ERPS2a、ERPS3,二者的单糖组成均为甘露糖、鼠李糖、半乳糖醛酸、葡萄糖、半乳糖、阿拉伯糖,ERPS2a的单糖物质的量比为2.1∶2.6∶3.4∶1.0∶7.9∶4.9,ERPS3的单糖物质的量比为1.0∶17.6∶2.7∶3.2∶19.8∶9.4。结论吴茱萸多糖得到有效分离纯化,两纯组分所含单糖种类相同,仅各单糖的组成比例不同。
OBJECTIVE To extract, purify and purify polysaccharides from Evodia rutaecarpa to further study the monosaccharide composition of the main components of Evodia rutaecarpa. Methods Crude polysaccharides were extracted by water extraction and alcohol precipitation method, and bleached by hydrogen peroxide (H2O2) and deproteinized by Sevag method. The purified products were prepared by dialysis. DEAE-32 cellulose anion exchange column and Sephacryl S-400 SF gel column were used to separate and purify the polysaccharides. After the 1-phenyl-3-methyl-5-pyrazolone (PMP) pre-column derivatization HPLC analysis of monosaccharide composition. Results The two major polysaccharides, ERPS2a and ERPS3, were isolated and purified from Evodia rutaecarpa. Both of the monosaccharides were mannose, rhamnose, galacturonic acid, glucose, galactose, arabinose and monosaccharide of ERPS2a Is 2.1: 2.6: 3.4: 1.0: 7.9: 4.9, the amount ratio of monosaccharide material of ERPS3 is 1.0: 17.6: 2.7: 3.2: 19.8: 9.4. Conclusion Fructus Evodia polysaccharide was effectively isolated and purified, the two pure components contained the same type of monosaccharides, only the composition of the proportion of different monosaccharides.
