目的探讨Ⅱ型跨膜丝氨酸蛋白酶4(TMPRSS4)和上皮型钙黏蛋白(E-cadherin)在胰腺导管腺癌组织中的表达及两者相关性以及其临床意义。方法 Western blot法检测16例胰腺导管腺癌和配对癌旁组织TMPRSS4和E-cadherin蛋白表达情况;免疫组织化学方法检测54例胰腺导管腺癌标本和4例正常胰腺组织中TMPRSS4和E-cadherin蛋白表达情况。结果胰腺导管腺癌组织TMPRSS4蛋白表达明显高于配对癌旁组织,E-cadherin蛋白表达癌旁组织明显高于癌组织(P<0.01);TMPRSS4蛋白阳性表达和E-cadherin表达缺失与胰腺导管腺癌分化程度、淋巴结移、临床分期相关(P<0.05);TMPRSS4和E-cadherin蛋白表达呈负相关(r=-0.419,P=0.002)。结论 TMPRSS4及E-cadherin在胰腺导管腺癌中的表达缺失与肿瘤恶性程度密切相关,联合检测2种蛋白有助于提高胰腺癌恶性程度的判断。
Objective To investigate the expression of type Ⅱ transmembrane serine protease 4 (TMPRSS4) and E-cadherin in pancreatic ductal adenocarcinoma and their relationship and their clinical significance. Methods The protein expression of TMPRSS4 and E-cadherin in 16 cases of pancreatic ductal adenocarcinoma and matched paracancerous tissues was detected by Western blot. The expressions of TMPRSS4 and E-cadherin in 54 cases of pancreatic ductal adenocarcinoma and 4 cases of normal pancreatic tissue were detected by immunohistochemistry Express the situation. Results The expression of TMPRSS4 protein in pancreatic ductal adenocarcinoma was significantly higher than that in adjacent tissues and the expression of E-cadherin was significantly higher in paracancer tissues (P <0.01). The positive expression of TMPRSS4 and the loss of E-cadherin in pancreatic ductal adenocarcinoma (P <0.05). There was a negative correlation between the expression of TMPRSS4 and E-cadherin protein (r = -0.419, P = 0.002). Conclusion The lack of expression of TMPRSS4 and E-cadherin in pancreatic ductal adenocarcinoma is closely related to the degree of malignancy of the tumor. Combined detection of the two proteins can help to improve the judgment of the malignant degree of pancreatic cancer.
