目的:观察肾衰养真胶囊(Shenshuai Yangzhen Capsule,SYC)对慢性肾衰竭(Chronic renal failure,CRF)营养不良大鼠下丘脑信号传导转录活化因子3(Signal transducer and activator of transcription3,STAT3)表达的影响。方法:采用含0.5%腺嘌呤及4%酪蛋白饲料联合喂养的方法制作CRF营养不良大鼠模型,检测BUN、Scr、Hb,Alb等生化指标,观察其营养不良发生时间,符合CRF营养不良模型的,随机分为正常组、模型组、SYC组和开同组。治疗4周后,应用放射免疫法检测血浆瘦素(Leptin,LP)水平;Western免疫印迹法检测下丘脑STAT3蛋白表达水平。结果:治疗后,SYC组大鼠营养状况明显改善Hb、Alb显著升高,血浆LP及下丘脑STAT3蛋白表达显著下调。结论:SYC可改善CRF营养不良大鼠的营养状况,其机制可能是通过下调血浆LP水平及其信号转导蛋白STAT3表达来实现的。
Objective: To observe the effect of Shenshuai Yangzhen Capsule (SYC) on the expression of Signal transducer and activator of transcription3 (STAT3) in Chronic renal failure (CRF) malnutrition rats. influences. Methods: The CRF malnutrition rat model was made by feeding 0.5% adenine and 4% casein feed. The biochemical indexes such as BUN, Scr, Hb and Alb were measured to observe the time of malnutrition, which was consistent with CRF malnutrition model Were randomly divided into normal group, model group, SYC group and open group. After 4 weeks of treatment, the level of plasma leptin (LP) was detected by radioimmunoassay. The expression of STAT3 protein in hypothalamus was detected by Western immunoblotting. Results: After treatment, the nutrition status of rats in SYC group was significantly improved, Hb and Alb were significantly increased, and the expression of LP and STAT3 protein in hypothalamus were significantly down-regulated. Conclusion: SYC can improve the nutritional status of rats with CRF malnutrition, and its mechanism may be through down-regulation of plasma LP and expression of signal transducer and transmitter STAT3.
