目的探讨P53基因在结肠癌组织中的表达水平,研究其与结肠癌发生、发展的关系。方法建立实时荧光定量PCR方法,检测各组织中P53基因的表达情况。结果扩增曲线呈典型的“S”型曲线;熔解曲线分析扩增产物显示单一的峰,熔解温度(Tm)为60℃。对照组中P53基因的表达明显高于癌组和癌旁组(P<0.05);癌组中P53基因的表达明显低于癌旁组(P<0.05)。结论实时荧光定量PCR方法具有高敏感性和高特异性等优点,可作为进一步研究P53基因的方法;p53基因的表达量可以作为判断结肠癌患者预后的重要指标。
Objective To investigate the expression of P53 gene in colon cancer and its relationship with the occurrence and development of colon cancer. Methods Real-time quantitative PCR method was established to detect the expression of P53 gene in various tissues. Results The amplification curve showed a typical “S” curve. The melting curve analysis showed a single peak and the melting temperature (Tm) was 60 ℃. The expression of P53 gene in the control group was significantly higher than that in the cancer group and adjacent non-cancerous group (P <0.05). The expression of P53 gene in the cancer group was significantly lower than that in the adjacent non-cancerous group (P <0.05). Conclusion Real-time PCR has the advantages of high sensitivity and specificity, which can be used as a method to further study P53 gene. The expression of p53 gene can be used as an important index to judge the prognosis of patients with colon cancer.
