本文介绍5 F U 持续静脉滴注及小剂量顺铂联合化疗方案( F P 方案) 。5 F U 代谢产物磷酸氟尿嘧啶脱氧核苷酸、还原型四氢叶酸及胸腺嘧啶脱氧核苷结合成三重复合体,阻碍肿瘤细胞 D N A 的合成,而小剂量 D D P 能够通过与细胞膜上蛋氨酸转运蛋白的结合而阻止蛋氨酸进入细胞内,使细胞内还原型四氢叶酸增加,从而间接破坏肿瘤细胞 D N A 的合成,这种作用即生物化学调节作用。此方案对恶性肿瘤尤其是胰腺癌有较高疗效。
This article describes the 5 F U continuous intravenous infusion and low dose cisplatin combined chemotherapy (F P program). The 5 F U metabolites, fluorouracil phosphate deoxynucleotide, reduced tetrahydrofolate and thymidine, combine to form a triple complex, impeding the synthesis of D N A in tumor cells, while the low-dose D D P can pass through the cell membrane. The combination of the methionine transporter prevents the entry of methionine into the cell, increases the intracellular reduced tetrahydrofolate, and indirectly destroys the synthesis of D N A in the tumor cells. This effect is biochemical regulation. This program has a higher effect on malignant tumors, especially pancreatic cancer.
