目的:测定不同原产地半夏块茎中半夏凝集素(PTA)的含量。方法:采用聚丙烯酰胺凝胶电泳法对38个半夏居群块茎中的PTA进行分离,并用凝胶成像系统定量分析PTA的含量。结果:不同居群间PTA含量差异较大,每克干燥块茎中的PTA含量介于0.0249~0.0662g之间。PTA含量大于0.05g的高含量居群占50%;低于0.03g的低含量居群占5%;介于0.03g和0.05g之间的中等含量居群占45%。秦岭淮河一带的居群PTA含量普遍很高,四川省的高含量半夏居群较少,且仅有的四个高含量居群全分布在沱江沿岸地区。结论:PTA的含量受基因型控制,与半夏居群原产地有关。
Objective: To determine the content of PTA in tubers of Pinellia ternata from different origins. Methods: Polyacrylamide gel electrophoresis (PAGE) was used to separate PTA in tubers from 38 populations of Pinellia ternata, and the content of PTA was quantitatively analyzed by gel imaging system. Results: The content of PTA in different populations was quite different. The content of PTA in the dry tubers per gram ranged from 0.0249 to 0.0662g. 50% of high-level populations with PTA> 0.05g, 5% of low-level populations below 0.03g, and 45% of middle-level populations between 0.03g and 0.05g. The population of PTA in the Huaihe River in Qinling Mountains is generally high. The populations of high-level pinellia in Sichuan Province are few, and the only four high-content populations are located in the Tuojiang River. Conclusion: The content of PTA is controlled by genotype, which is related to the origin of Pinellia population.
