目的:优化阿里红中总三萜酸的提取工艺,并建立其含量测定方法。方法:以齐墩果酸为对照品,采用分光光度法测定三萜酸含量;采用单因素试验考察提取溶剂种类及浓度、料液比、温度、提取时间及提取次数对总三萜酸得率的影响,在此基础上,采用L9(34)正交试验优选阿里红中总三萜酸提取工艺。结果:最佳提取工艺为30倍量90%乙醇于60℃超声提取3次,每次30 min。在此条件下,阿里红中总三萜酸质量分数35.1%。各因素影响次序为提取温度>液料比>提取时间>乙醇体积分数。结论:阿里红中三萜酸含量很高,该优选工艺稳定可行,可推广于大生产。
Objective: To optimize the extraction process of total triterpene acid in Ali red and to establish its determination method. Methods: Oleanolic acid was used as reference substance, and the content of triterpenic acid was determined by spectrophotometry. The single factor test was used to investigate the effects of extraction solvent type and concentration, solid-liquid ratio, temperature, extraction time and extraction times on total triterpene acid yield On the basis of this, orthogonal experiment of L9 (34) was used to optimize the extraction process of total triterpene acid in Ali red. Results: The best extraction process was 30 times the amount of 90% ethanol at 60 ℃ ultrasonic extraction three times, each time 30 min. Under this condition, the total triterpene acid content of Ali red 35.1%. The order of influence of each factor is extraction temperature> liquid material ratio> extraction time> ethanol volume fraction. Conclusion: The content of triterpenic acid in Ali red is very high. The optimal process is stable and feasible and can be extended to large-scale production.
