研究骨髓间充质干细胞(BMSC)对同种异体T淋巴细胞不同亚群的细胞因子分泌、细胞周期和增殖能力的影响,探讨BMSC对同种异体T淋巴细胞各亚群功能的影响,以阐明BMSC对免疫调节的作用机制。分离纯化鉴定BALB/c小鼠BMSC,取C57小鼠脾脏制备淋巴细胞悬液,免疫磁珠法富集T淋巴细胞,BMSC与T淋巴细胞共培养,监测淋巴细胞增殖能力,流式细胞术分析CD4+、CD8+、CD4+CD45RBhigh、CD4+CD45RBlow、CD8+CD45RBhigh、CD8+CD45RBlow各T淋巴细胞亚群IFN-γ、IL-4分泌水平及对DNA周期的影响。结果BMSC能明显抑制由同种异体抗原诱导的T淋巴细胞增殖,可使CD4+和CD8+T淋巴细胞分泌IFN-γ水平下降,IL-4水平无影响。具有明显抑制CD4+CD45RBhigh、CD4+CD45RBlow和CD8+CD45RBhigh、CD8+CD45RBlow亚群IFN-γ的表达并且在不同亚群抑制程度不同。骨髓间充质干细胞通过抑制T淋巴细胞的增殖和不同功能亚群分泌细胞因子从而抑制免疫反应。
To investigate the effect of BMSC on cytokine secretion, cell cycle and proliferation in different subpopulations of allogeneic T lymphocytes and to explore the effect of BMSC on the function of allogeneic T lymphocyte subsets in order to clarify Mechanism of BMSC on immune regulation. Isolation and purification of BALB / c mouse BMSC, C57 mouse spleen lymphocyte suspension was prepared, immunomagnetic beads enrich T lymphocytes, BMSC and T lymphocytes were co-cultured to monitor lymphocyte proliferation, flow cytometry analysis CD4 +, CD8 +, CD4 + CD45RBhigh, CD4 + CD45RBlow, CD8 + CD45RBhigh, CD8 + CD45RBlow T-lymphocyte subsets IFN-γ, IL-4 secretion and DNA cycle. Results BMSC could significantly inhibit the proliferation of T lymphocytes induced by alloantigen, which could decrease the level of IFN-γ secreted by CD4 + and CD8 + T lymphocytes and had no effect on the level of IL-4. With significant inhibition of the expression of IFN-γ of CD4 + CD45RBhigh, CD4 + CD45RBlow and CD8 + CD45RBhigh, CD8 + CD45RBlow subsets and different levels of inhibition in different subsets. Bone marrow mesenchymal stem cells suppress immune response by inhibiting the proliferation of T lymphocytes and secretion of cytokines by different functional subpopulations.
