目的研究加贝酯对D-氨基半乳糖(GaIN)加脂多糖(LPS)诱导的大鼠急性肝衰竭的保护作用。方法在建立GaIN+LPS诱导急性肝衰竭小鼠模型的基础上,用分光光度法检测血清中ALT、AST水平和肝匀浆MDA、GSH-PX、SOD含量;ELISA法检测血清中TNF-α、IL-1β和IL-6水平;HE染色法对肝脏组织作病理检查。结果加贝酯(25 mg·kg~(-1)、50 mg·kg~(-1)、100 mg·kg~(-1))腹腔注射给药可明显降低肝损伤小鼠血清ALT、AST活性,同时能减少肝匀浆MDA含量,但对肝匀浆GSH-px、SOD活性无明显影响;病理检查也发现其可明显减轻肝组织坏死范围及程度,减少炎细胞浸润。加贝酯可明显抑制肝衰竭小鼠血清TNF-α、IL-1β和IL-6的产生。结论加贝酯对GaIN+LPS诱导的急性肝功能衰竭大鼠具有保护作用。
Objective To study the protective effect of Gabapeptide against acute hepatic failure induced by L-galactosamine (GaIN) and lipopolysaccharide (LPS) in rats. Methods Serum levels of ALT and AST and the levels of MDA, GSH-PX and SOD in liver homogenates were determined by establishing a model of acute liver failure induced by GaIN + LPS. Serum TNF-α, IL-1β and IL-6 levels; HE staining of liver tissue for pathological examination. Results Gabapeptide (25 mg · kg -1, 50 mg · kg -1, 100 mg · kg -1) could significantly reduce the serum levels of ALT and AST Activity, at the same time reduce the content of MDA in liver homogenate, but had no significant effect on the activity of GSH-px and SOD in liver homogenate. Pathological examination also found that it could obviously reduce the scope and extent of liver necrosis and reduce inflammatory cell infiltration. Gabapeptide significantly inhibited the production of TNF-α, IL-1β and IL-6 in serum of mice with liver failure. Conclusion Gabapeptide has a protective effect on GaIN + LPS-induced acute liver failure in rats.
