目的探讨甲基苄基亚硝胺(MBNA)致食管癌前病变时大鼠血清氧化还原水平的变化及鸦胆子油的影响。方法Wistar大鼠按3.5mg/kg皮下注射MBNA溶液,每周2次,维生素C组自诱变首日起皮下注射,每周5次,连续10周,治疗4周后处死所有大鼠,检测总SOD活性、总谷胱甘肽水平和总抗氧化能力并观察食管黏膜变化。结果各组瘤体数较模型组有所减少(P<0.01),除维生素C组外,各组瘤体体积均无差异(P>0.05);模型组较正常组总SOD活性,总谷胱甘肽水平和总抗氧化能力有所升高(P<0.05);鸦胆子油组较模型组降低(P<0.01)。结论经鸦胆子油治疗后,总SOD活性,总谷胱甘肽水平和总抗氧化能力降低,增强肿瘤组织的氧化损伤可能是鸦胆子抗肿瘤的机制之一。
Objective To investigate the changes of serum redox levels in rats with methylprednisolone (MBNA) induced esophageal precancerous lesions and the effects of Brucea javanica oil. Methods Wistar rats were injected subcutaneously with 3.5 mg / kg of MBNA solution twice a week. The vitamin C group was injected subcutaneously on the first day of the mutagenesis five times a week for 10 weeks. After 4 weeks of treatment, all the rats were killed and tested Total SOD activity, total glutathione level and total antioxidant capacity and observe esophageal mucosal changes. Results Compared with the model group, the number of tumor in each group was decreased (P <0.01). There was no difference in tumor volume among groups except vitamin C group (P> 0.05) The level of glycans and the total antioxidant capacity increased (P <0.05). Brucea javanica oil group was lower than model group (P <0.01). Conclusion After treatment with Brucea javanica oil, the total SOD activity, total glutathione level and total antioxidant capacity decreased. Increasing the oxidative damage of tumor tissue may be one of the mechanisms of Brucea javanica anti-tumor.
