目的通过建立人LOVO细胞结肠癌裸鼠皮下移植瘤模型,研究血管生成抑制剂(TNP-470)联合分化诱导剂(RA)对结肠癌细胞凋亡的影响。方法建立人LOVO细胞结肠癌裸鼠皮下移植瘤模型,随机分为4组。模型建立后分组治疗,至治疗结束时处死裸鼠,检测肿瘤组织结肠癌细胞凋亡指数(AI)。结果治疗组与对照组癌细胞AI相比,差异有统计学意义(P<0.05);且RA+TNP-470组与RA组和TNP-470组癌细胞的AI相比,差异也有统计学意义(P<0.05)。联合用药组肿瘤组织中可见较多的凋亡细胞。结论血管生成抑制剂TNP-470与分化诱导剂RA联合治疗结肠癌裸鼠皮下移植瘤比TNP-470或RA单药治疗具有更强的抑瘤作用,肿瘤组织结肠癌凋亡细胞明显增加。
OBJECTIVE: To study the effect of angiogenesis inhibitor (TNP-470) combined with differentiation inducer (RA) on the apoptosis of colon cancer cells by establishing a subcutaneous xenograft model of human LOVO colon cancer in nude mice. Methods Human ovariectomized xenograft model of human LOVO colon cancer was established and randomly divided into 4 groups. After the model was established, the rats were sacrificed and the nude mice were sacrificed at the end of treatment to detect the apoptosis index (AI) of the colon cancer cells. Results The AI of cancer cells in the treatment group and the control group was significantly different (P <0.05). There was also a significant difference in AI between the RA + TNP-470 group and the RA and TNP-470 groups (P <0.05). More apoptotic cells were found in tumor tissues of combination group. Conclusion The combination of angiogenesis inhibitor TNP-470 and differentiation inducer RA in the treatment of subcutaneously transplanted colon cancer in nude mice has a stronger anti-tumor effect than that of TNP-470 or RA alone. The apoptotic cells in colon cancer tissues are significantly increased.
