目的:建立酚试剂分光光度法检测亚单位流感疫苗中甲醛含量的方法。方法:采用酚试剂分光光度法,通过改变酚试剂,盐酸,硫酸铁铵浓度,反应时间优化检测条件。结果:在反应体系5 mL中,样本500μL与0.85 mg.mL-1酚试剂2 mL35℃反应10 min后,加入0.1 mol.L-1盐酸2mL和1%硫酸铁铵0.5 mL,35℃反应20 min为最佳反应条件,在0.1~5μg.mL-1范围内呈线性相关,样品加标回收率为98.5%。结论:该方法适用于实验室对亚单位流感疫苗中微量甲醛含量的常规检测。
Objective: To establish a method for the determination of formaldehyde in subunit influenza vaccine by phenol reagent spectrophotometry. Methods: Phenol reagent spectrophotometry was adopted to optimize the test conditions by changing the concentration of phenol reagent, hydrochloric acid, ferric ammonium sulfate and reaction time. Results: In 5 mL of the reaction system, 500 μL of the sample was reacted with 2 mL of phenol of 0.85 mg.mL-1 at 35 ℃ for 10 min, and then 2 mL of 0.1 mol·L-1 hydrochloric acid and 0.5 mL of 1% ammonium ferric sulfate were added. min was the best reaction condition, linearly correlated in the range of 0.1 ~ 5μg.mL-1, the recoveries of spiked samples were 98.5%. Conclusion: This method is suitable for the routine laboratory detection of trace formaldehyde in the subunit influenza vaccine.
