目的:设计正交试验,探讨水提醇析法提取、分离纯化辛夷花多糖的最佳工艺。方法:按正交试验设计方案,设定对提取率和含量影响较大的4因素即提取温度、时间、乙醇浓度和氯仿萃取次数,各因素设定3水平,研究辛夷花多糖提取的最佳工艺。辛夷花多糖的百分含量以硫酸-苯酚法测定。结果:按正交试验方案进行试验,经过计算和综合分析,得出辛夷花多糖的最佳提取工艺为水浸提2h,浸提温度90℃,氯仿萃取2次,沉析用乙醇浓度为80%。结论:该优选工艺可用于提取辛夷花多糖,且所得辛夷花多糖含量较高。
OBJECTIVE: To design an orthogonal test to explore the optimum process for extracting, separating and purifying Polysaccharide from the flower of Magnolia officinalis. Methods: According to the orthogonal experimental design scheme, the 4 factors that have a great influence on the extraction rate and content are the extraction temperature, time, ethanol concentration and chloroform extraction times. Each factor is set to 3 levels, and the optimal extraction of polysaccharide from magnolia flower is studied. Process. The percentage of the polysaccharides of Magnolia flower was determined by sulfuric acid-phenol method. Results: According to the orthogonal test scheme, after the calculation and comprehensive analysis, it was concluded that the best extraction process of Polygonatum sinensis was water extraction for 2 hours, extraction temperature of 90°C, and chloroform extraction for 2 times. The concentration of ethanol for the coagulation was 80. %. Conclusion: The optimized process can be used to extract the polysaccharides from M. chinensis and the content of polysaccharides obtained from M. chinensis is higher.
