目的:探讨肾炎Ⅱ号水丸(SY)对紫癜性肾炎(HSPN)小鼠血清IgA-纤维连接蛋白聚合物(IgA-FN)含量的影响。方法:采用口服麦胶蛋白+尾静脉注射印度墨水的方法建立小鼠紫癜性肾炎(HSPN)模型。灌胃给药17周后,ELISA法测定各组小鼠血清IgA-FN含量;免疫荧光法检测各组小鼠肾组织IgA沉积。结果:与空白组比较,模型组IgA呈团块状在系膜区沉积明显增多(P<0.01);与模型组比较,SYH组IgA沉积明显减少(P<0.01),SYM组IgA沉积明显减少(P<0.05)。与空白组比较,模型组血清IgA-FN含量明显升高(P<0.01);与模型组比较,实验各组皆可以明显降低模型小鼠血清IgA-FN的含量(P<0.01)。结论:SY降低HSPN模型血清中IgA-FN表达从而促进系膜区IgA的清除可能是其防治HSPN的机理之一。
Objective: To investigate the effect of nephritis Ⅱ water pill (SY) on serum IgA-fibronectin (IgA-FN) in purpura nephritis (HSPN) mice. Methods: Murine purpura nephritis (HSPN) model was established by injecting Indian ink with gliadin and tail vein. Serum IgA-FN levels were measured by ELISA in 17 weeks after intragastric administration. IgA deposition in renal tissues was detected by immunofluorescence. Results: Compared with the blank group, IgA deposition in the model group was significantly increased in the mesangial area (P <0.01). Compared with the model group, the IgA deposition in the SYH group was significantly decreased (P <0.01), and the IgA deposition in the SYM group was significantly decreased (P <0.05). Compared with the blank group, the content of serum IgA-FN in the model group was significantly increased (P <0.01). Compared with the model group, the content of IgA-FN in the model group was significantly decreased (P <0.01). CONCLUSIONS: SY may reduce IgA-FN expression in serum of HSPN model and promote the clearance of IgA in mesangial area, which may be one of the mechanisms of preventing and treating HSPN.
