目的:研究不同程度PEG胁迫对黄芩悬浮细胞中黄芩苷、黄芩素的积累及相关基因表达的影响。方法:采用HPLC法检测黄芩悬浮细胞中黄芩苷、黄芩素的含量,用酶标仪检测细胞内游离脯氨酸含量,并用半定量RT-PCR的方法对相关基因的表达进行分析。结果与结论:黄芩悬浮细胞内游离脯氨酸含量随PEG胁迫浓度升高而升高,10%PEG胁迫黄芩悬浮细胞可以模拟干旱条件,提高悬浮细胞PAL基因表达量且会促进黄芩素的积累。20%PEG胁迫提高了UBGAT基因的表达量,但同时APX酶活性的提高抑制了黄芩素参与清除活性氧,导致了黄芩素向黄芩苷的转化。
Objective: To study the effects of PEG stress on the accumulation of baicalin and baicalein and their related genes in Scutellaria baicalensis suspension cells. Methods: The content of baicalin and baicalein in suspension cells of Scutellaria baicalensis Georgi was detected by HPLC. The content of free proline in cells was detected by microplate reader. The expression of related genes was analyzed by semi-quantitative RT-PCR. RESULTS AND CONCLUSION: The contents of free proline in suspension cells of Scutellaria baicalensis Georgi increased with the increase of PEG stress concentration. Scutellaria baicalensis suspension cells with 10% PEG stress could simulate the drought conditions, increase the PAL gene expression in suspension cells and promote the accumulation of baicalein. 20% PEG increased the expression of UBGAT gene, but at the same time, the increase of APX enzyme activity inhibited the participation of baicalein in scavenging reactive oxygen species, resulting in the conversion of baicalein to baicalin.
