目的:研究天丝饮对去势所致老年痴呆模型行为学及神经营养相关的研究。方法:50只SD大鼠随机分为正常组、模型组、假手术组、雌二醇组、天丝饮组。通过双侧去卵巢造成老年痴呆模型,手术1周后,雌二醇组及天丝饮组分别给予雌二醇(0.2mg/m L)、天丝饮(0.35g/kg),灌胃8周。采用Morris水迷宫检测其5d的学习行为能力,Western blot测试其海马组织中p-p38 MAPK,p-CREB,BDNF的含量。结果:在Morris水迷宫中,与模型组比较,天丝饮组潜伏距离缩短(P<0.05),穿越平台次数明显增多(P<0.01)。Western blot表明,与模型组比较,天丝饮组能降低大鼠大脑海马区p-p38 MAPK的表达,升高p-CREB、BDNF的表达(P<0.01)。结论:天丝饮在一定程度上通过抑制大鼠大脑海马神经区神经凋亡和增强神经营养,改善去势大鼠的学习记忆能力。
OBJECTIVE: To study the effect of Tencel on the behavioral and neurotrophic effects of castrated dementia model. Methods: Fifty SD rats were randomly divided into normal group, model group, sham operation group, estradiol group and Tencel drink group. The model of Alzheimer’s disease was established by bilateral ovariectomy. Estradiol (0.2mg / m L), Tencel (0.35g / kg), gavage 8 week. Morris water maze was used to detect the learning behavior of rats for 5 days. The contents of p-p38 MAPK, p-CREB and BDNF in hippocampus were detected by Western blot. Results: In the Morris water maze, compared with the model group, the latency of Tencel Drink Group was shorter (P <0.05) and the number of crosses was significantly increased (P <0.01). Western blot showed that compared with the model group, Tiansiyin Decoction could reduce the expression of p-p38 MAPK and increase the expression of p-CREB and BDNF in hippocampus of rats (P <0.01). Conclusion: Tencel can improve the learning and memory of ovariectomized rats by inhibiting neuronal apoptosis and enhancing neurotrophic activity in rat hippocampus to a certain extent.
