目的:通过观察龟鹿二仙胶及其拆方对豚鼠软骨细胞Ⅱ型胶原和蛋白多糖合成的影响,探讨龟鹿二仙胶的配伍机制。方法:体外培养豚鼠膝关节软骨细胞,免疫组化染色检测龟鹿二仙胶及其拆方对软骨细胞Ⅱ型胶原合成的影响,Alcian Blue染色检测软骨细胞蛋白多糖合成。结果:龟鹿二仙胶促进软骨细胞Ⅱ型胶原和蛋白多糖表达极显著(P<0.01),龟板和鹿角均能显著提高Ⅱ型胶原和蛋白多糖表达(P<0.05),但药效显著低于龟鹿二仙胶(P<0.05);人参和枸杞对Ⅱ型胶原和蛋白多糖表达均无明显促进作用。结论:龟鹿二仙胶可显著促进软骨细胞Ⅱ型胶原和蛋白多糖的表达,这是其保护软骨细胞,延缓软骨退变的作用机制之一。龟鹿二仙胶发挥此作用的主要药物是龟板和鹿角;人参和枸杞本身并不能促进Ⅱ型胶原和蛋白多糖表达,而与龟板和鹿角配合,可明显增强龟板和鹿角的促进软骨细胞Ⅱ型胶原和蛋白多糖表达的作用。
OBJECTIVE: To observe the effect of the turtle deer glue and its decomposed formulas on the synthesis of type Ⅱ collagen and proteoglycan of guinea pig chondrocytes and to explore the compatibility mechanism of the two insects. Methods: The chondrocytes of guinea pigs were cultured in vitro. The effects of Guilu Erxian and its decomposed formulas on the synthesis of type Ⅱ collagen in chondrocytes were detected by immunohistochemical staining. The synthesis of proteoglycan in chondrocytes was detected by Alcian Blue staining. Results: The expression of type Ⅱ collagen and proteoglycan in chondrocytes was significantly enhanced by Guilu Erxian (P <0.01), and the expression of type Ⅱ collagen and proteoglycan was significantly increased in turtle shell and antler (P <0.05), but the effect was significantly lower (P <0.05). Ginseng and Lycium did not significantly promote the expression of type Ⅱ collagen and proteoglycan. Conclusion: Guilu Erxian gum can significantly promote the expression of type Ⅱ collagen and proteoglycan of chondrocytes, which is one of the mechanisms of its protection of chondrocytes and delaying the degeneration of cartilage. Turtle deer and cents glue play the role of the main drugs are turtle shell and antlers; ginseng and Chinese wolfberry itself can not promote the expression of type Ⅱ collagen and proteoglycans, but with the turtle shell and antlers, can significantly enhance the turtle shell and antlers promote cartilage type Ⅱ The role of collagen and proteoglycan expression.
