目的 :为观察tPNS对HCS刺激的RASMC增殖及其释放NO的影响。方法 :运用兔HCS刺激体外培养的RASMC而形成的细胞异常增殖为模型 ,通过MTT法、电镜观察和Green法 ,观察了tPNS对RASMC的保护作用及其对NO释放的影响。结果 :HCS可明显抑制NO的释放、刺激RASMC增殖并导致其表型的改变 ,而tPNS可诱导NO的释放 ,明显抑制HCS对RASMC的刺激增殖作用 ,与HCS组相比具有显著差异 (P <0 .0 1)。结论 :tPNS抗动脉粥样硬化作用可能部分是通过调节NO的合成而抑制RASMC增殖来实现的。
OBJECTIVE: To observe the effect of tPNS on proliferation and release of NO in RASMC stimulated by HCS. Methods: Rabbit HCS stimulated RASMC proliferation in vitro. The effects of tPNS on RASMC and its effect on NO release were observed by MTT assay, electron microscopy and Green assay. Results: HCS significantly inhibited the release of NO, stimulated the proliferation of RASMC and resulted in its phenotype change. However, tPNS could induce the release of NO and significantly inhibit the proliferation of RASMC induced by HCS, which was significantly different from HCS group (P < 0 .0 1). CONCLUSION: Anti-atherosclerotic effects of tPNS may be partially mediated by the inhibition of RASMC proliferation by regulating NO synthesis.
