目的建立同时测定咳喘胶囊(I)中绿原酸与黄芩苷的高效液相色谱(HPLC)法。方法采用双波长HPLC法,以甲醇为流动相A,0.4%磷酸溶液为流动相B,梯度洗脱;检测波长为327 nm(绿原酸)、280 nm(黄芩苷),柱温40℃,体积流量1.0 m L/min,进样量,10μL。结果绿原酸在6.29~41.96μg线性良好(r=0.999 9,n=5),黄芩苷在5.78~38.52μg线性良好(r=0.999 9,n=5),平均回收率分别为99.7%和100.0%,RSD分别为0.9%和0.9%。结论本法操作简便,重复性好,可有效控制咳喘胶囊的质量。
Objective To establish a HPLC method for the simultaneous determination of chlorogenic acid and baicalin in Kechuan Capsule (I). Methods Two-wavelength HPLC with methanol as mobile phase A and 0.4% phosphoric acid as mobile phase B was used for the gradient elution. The detection wavelength was 327 nm (chlorogenic acid), 280 nm (baicalin) Volume flow 1.0 m L / min, injection volume, 10 μL. Results The linearity of chlorogenic acid was good at 6.29 ~ 41.96μg (r = 0.999 9, n = 5). The average recovery of baicalin was 5.78 ~ 38.52μg (r = 0.999 9, n = 5) 100.0% with RSD of 0.9% and 0.9% respectively. Conclusion This method is simple, reproducible, and can effectively control the quality of Kechuan capsule.
