目的探讨黄芪注射液治疗小鼠柯萨奇B3致病毒性心肌炎(VMC)的疗效及其机制。方法Balb/C鼠24只腹腔感染柯萨奇病毒B3亲心肌细胞株后,随机等分为两组:治疗组(黄芪注射液每天10g/kg腹腔注射)和对照组。实验第8天取心肌和血液标本,进行心肌病理检查,用逆转录聚合酶链反应分析心肌穿孔素(PFP)和Fas配体(FasL)mRNA表达水平及进行黄芪对其影响的相关性分析。结果 (1)治疗组心肌组织的病理改变明显轻于对照组;(2)治疗组心肌PFP mRNA的表达明显低于对照组(P<0.01);(3)心肌FasL mRNA表达水平治疗组明显高于对照组(P<0.01);(4)直线回归与相关性分析显示心肌PFP mRNA与FasL mRNA水平呈显著负相关(r=-0.78,P<0.01)。结论 黄芪注射液能影响VMC中PFP介导的细胞毒性作用,减轻心肌损伤和炎症反应,对VMC有明显的治疗作用。
Objective To investigate the efficacy and mechanism of Huangqi injection in treating Coxsackie B3 induced viral myocarditis (VMC) in mice. Methods After intraperitoneal infection with Coxsackie virus B3 myocardiomyocytes in 24 Balb/C mice, they were randomly divided into two groups: the treatment group (intraperitoneal injection of Huangqi injection at 10g/kg daily) and the control group. Myocardial and blood samples were collected on the 8th day of the experiment for myocardial pathological examination. The expression of perforin (PFP) and FasL (FasL) mRNA was analyzed by reverse transcription polymerase chain reaction and correlation analysis was performed on the effects of Astragalus membranaceus on it. Results (1) The pathological changes of myocardial tissue in the treated group were significantly lighter than those in the control group; (2) The expression of PFP mRNA in the treated group was significantly lower than that in the control group (P<0.01); (3) The expression level of myocardial FasL mRNA was significantly higher in the treated group In the control group (P <0.01); (4) linear regression and correlation analysis showed a significant negative correlation between myocardial PFP mRNA and FasL mRNA levels (r =- 0.78, P <0.01). Conclusion Astragalus injection can affect the PFP-mediated cytotoxicity in VMC, reduce myocardial injury and inflammatory response, and have a significant therapeutic effect on VMC.
