目的:观察镇肝熄风汤对动物模型的性激素水平(FSH、LH、E2)及胸主动脉雌激素受体(Estrogen receptor,ER)的影响。方法:建立动物模型,设置对照组、模型组、雌激素组、镇肝熄风汤组,治疗4周后,对各组的FSH、LH、E2、ER等指标进行检测。结果:①去势后模型组较对照组血清E2含量下降(P<0.05),镇肝熄风汤组较模型组血清E2含量上升(P<0.05);②大鼠血清FSH、LH含量过低,FSH<0.05IU/L,LH<0.07IU/L,无法比较;③模型组与对照组相比,ER下降,镇肝熄风汤组与雌激素组ER水平均升高(P<0.05)。结论:镇肝熄风汤可提高去势大鼠E2水平,具有调节生殖内分泌的功能,同时可提高胸主动脉胞浆ER水平。提示镇肝熄风汤治疗绝经综合征和抗动脉粥样硬化的机制可能是通过提高雌激素的数量及雌激素受体的表达,加强其靶器官和组织的生物学效应。
Objective: To observe the effects of Zhengan Xifeng Decoction on the levels of sex hormones (FSH, LH, E2) and estrogen receptor (ER) in thoracic aorta. Methods: Animal models were established. Control group, model group, estrogen group and Zhengan Xifeng Decoction group were set up. After 4 weeks of treatment, the indexes of FSH, LH, E2, and ER in each group were examined. RESULTS: 1 The serum E2 level in the model group was lower than that in the control group after castration (P<0.05). The serum E2 level in the Zhengan Xifeng Decoction group was higher than that in the model group (P<0.05); 2 The serum levels of FSH and LH in the rats were too low. FSH <0.05 IU/L, LH <0.07 IU/L, could not be compared; 3 ER decreased in model group and control group, ER levels in Zhengan Xifeng Decoction group and estrogen group increased (P<0.05) . Conclusion: Zhengan Xifeng Decoction can increase the level of E2 in ovariectomized rats, and it can regulate the function of reproductive endocrinology. At the same time, Zhengan Xifeng Decoction can increase the level of cytoplasmic ER in thoracic aorta. It is suggested that the mechanism of Zhengan Xifeng Decoction in treating menopause syndrome and anti-atherosclerosis may be to increase the number of estrogens and the expression of estrogen receptors, and to strengthen the biological effects of their target organs and tissues.
