目的建立一种灵敏,简便,快速测定人体血浆中尼莫地平血药浓度的HPLC方法。方法色谱柱:美国WatersSymmetry C18柱(5μm150mm×4.6mm);流动相:甲醇∶水∶三乙胺(65∶35∶0.02v/v/v);流速:1.0mL·min-1;检测波长:358nm,尼莫地平碱化后经正己烷/乙醚(1∶1v/v)提取后测试。结果尼莫地平在1.25~80μg.L-1范围内线性良好,尼莫地平与内标面积比为纵坐标进行线性回归,得回归方程为:y=0.0477x+0.0233,r2=0.9997,最低检测浓度为:1.25μg.L-1,方法回收率为:(96.44±8.49)%,(n=5),日内日间精密度为6.62%和10.90%;结论该方法适于人体血浆内尼莫地平的检测并可应用于药动学研究。
Objective To establish a sensitive, simple and rapid HPLC method for the determination of nimodipine in human plasma. Method Column: Waters Symmetry C18 column (5 μm 150 mm × 4.6 mm); mobile phase: methanol: water: triethylamine (65:35:0.02 v / v); flow rate: 1.0 mL · min -1; 358nm, nimodipine alkalized after n-hexane / ether (1: 1v / v) extraction test. Results The linearity of nimodipine in the range of 1.25 ~ 80μg.L-1 was good, and the linear regression of nimodipine and internal standard area ratio was the vertical axis. The regression equation was y = 0.0477x + 0.0233, r2 = 0.9997, The method recovery was (96.44 ± 8.49)%, (n = 5). The intra-day and inter-day precision was 6.62% and 10.90%, respectively. Conclusion The method is suitable for the determination of nimomo The detection of the horizon can be applied to pharmacokinetic studies.
