本研究利用转基因技术将直立型密穗基因dep1导入籼稻成熟胚愈伤组织中,以期获得直立穗型转基因水稻植株。结果显示已获得209个转基因再生植株,PCR检测结果表明阳性植株189株,阳性率90.4%;Southern Blot和RT-PCR检测结果表明目的基因已经整合到9R406基因组中,并且在转基因植株后代中能够稳定地遗传和表达。T1代转基因再生植株农艺性状考察结果表明,与对照相比,部分株系株高降低,穗粒数和实粒数增加,穗长变短,千粒重减小。
In this study, the erect vertical ear gene, dep1, was introduced into callus of mature embryo of indica rice by using transgenic technology in order to obtain erect panicle transgenic rice plants. The results showed that 209 transgenic plants had been obtained. The results of PCR showed that there were 189 positive plants, the positive rate was 90.4%. Southern Blot and RT-PCR results showed that the target gene had been integrated into 9R406 genome and could be stabilized in the progeny of transgenic plants Genetically and expressed. The results of the agronomic traits in the T1 generation transgenic plants showed that compared with the control, some plants had lower plant height, more grains per spike and real grain number, shorter spike length and lower 1000-grain weight.
