采用90只Wistar大鼠,以D半乳糖胺(Dgal)作为肝毒剂,建成急性肝损伤动物模型,筛选了最佳用药剂量,并研究了损伤后肝脏功能及组织形态的变化规律。结果表明,本实验条件下,Dgal450mg/kg体重为最适剂量,所致肝损伤组织形态变化显著且死亡率低(<24%),呈现的功能与形态变化程度一致,病变恒定。认为本模型可用于保肝新药的筛选与评价。
90 Wistar rats were used to treat D-gal as hepatotoxic agents to establish an animal model of acute liver injury. The optimal dose of D-gal was screened and the changes of hepatic function and histomorphology after injury were studied. The results showed that under the conditions of this experiment, D-gal450mg / kg body weight as the optimum dose, resulting in significant changes in liver injury tissue morphology and low mortality (<2 4%), showing the same degree of function and morphological changes, constant lesions . The model can be used for the screening and evaluation of new drugs for liver protection.
