目的探讨不同级别胶质瘤中Fas、Caspase-8的表达情况。方法针对68例经病理证实的人脑胶质瘤组织标本和15例正常对照标本,采用免疫组化SP法检测Fas、Caspase-8的表达。结果在低级别胶质瘤(LGG,Ⅰ~Ⅱ级)和高级别胶质瘤(HGG,Ⅲ~Ⅳ级)中,Fas阳性率分别为60.6%(20/33)和91.4%(32/35),Caspase-8的阳性率分别为81.8%(27/33)和48.6%(17/35)。胶质瘤病理分级越高,Fas的阳性率越高(χ2=8.968,P=0.003<0.05),Caspase-8的阳性率越低(χ2=8.221,P=0.004<0.05)。胶质瘤的恶性程度与Fas的表达呈正相关(rs=0.536,P<0.001),与Caspase-8的表达呈负相关(rs=-0.348,P<0.01)。结论胶质瘤细胞可对Fas系统诱导的凋亡产生耐受,Caspase-8功能的下降与胶质瘤的发生和发展密切相关。
Objective To investigate the expression of Fas and Caspase-8 in different grades of gliomas. Methods 68 cases of pathologically confirmed glioma tissue samples and 15 normal control specimens were detected by immunohistochemical SP method Fas, Caspase-8 expression. Results The positive rates of Fas were 60.6% (20/33) and 91.4% (32/35) in low grade gliomas (grade Ⅰ ~ Ⅱ) and high grade gliomas (grade Ⅲ ~ Ⅳ) ), The positive rates of Caspase-8 were 81.8% (27/33) and 48.6% (17/35), respectively. The higher the grade of glioma, the higher the positive rate of Fas (χ2 = 8.968, P = 0.003 <0.05) and the lower the positive rate of Caspase-8 (χ2 = 8.221, P = 0.004 <0.05). The malignant degree of glioma was positively correlated with the expression of Fas (rs = 0.536, P <0.001), but negatively correlated with the expression of Caspase-8 (rs = -0.348, P <0.01). Conclusion Glioma cells can tolerate apoptosis induced by Fas system. The decline of Caspase-8 function is closely related to the occurrence and development of glioma.
