目的:观察预热处理对运动后大鼠肝脏热休克蛋白(HSP)70表达和抗氧化能力的影响。方法:72只雄性Wistar大鼠随机分为9组:安静对照组(C)、热处理即刻组(H0)、热处理恢复24h组(H24)、预热处理恢复24h后再离心运动即刻组(HE0)、预热处理恢复24h后再离心运动后恢复24h组(HE24)、预热处理恢复24h后再离心运动恢复48h组(HE48)以及常温离心运动后即刻组(E0)、常温离心运动后24h组(E24)、常温离心运动后48h组(E48)。采用免疫印迹等方法对大鼠肝脏HSP70蛋白表达及SOD、MDA、GSH、GPT进行测定。结果:H0组大鼠肝脏HSP70蛋白表达水平为133%,与C组相比有增加趋势;H24组HSP70蛋白表达水平增加到175%,与C组相比有显著性差异(P<0.05)。E0组、E24组、E48组大鼠肝脏HSP70蛋白表达水平均显著高于C组(P<0.05),呈逐渐增高趋势。HE0组大鼠肝脏HSP70蛋白表达水平达到138%,也显著高于C组(P<0.05);但HE24组下降至89%,显著低于E24组(P<0.05);HE48组明显增加,达到224%,显著高于C组(P<0.05),亦高于E48组的176%,其差异具有统计学意义(P<0.05)。结论:预热处理明显使肝脏HSP70表达增加,并有延迟表达现象。预热处理会导致GSH明显下降,但对MDA和SOD影响较小或没有影响。预热处理不能缓解离心运动后GSH明显下降的趋势。
Objective: To observe the effects of preheating treatment on the expression of heat shock protein (HSP) 70 and the anti-oxidative capacity of rat liver after exercise. Methods: Seventy-two male Wistar rats were randomly divided into 9 groups: control group (C), heat treatment group (H0), heat treatment 24h group (H24), preheating treatment 24h after reperfusion group (HE0) , Preheated for 24 h, then resumed 24 h after centrifugation (HE 24), preheated for 24 h and then re-centrifuged for 48 h (HE 48), and immediately after centrifugation at room temperature (E 0) (E24), normal group at 48h after centrifugation (E48). Immunohistochemistry and other methods were used to detect HSP70 protein, SOD, MDA, GSH and GPT in rat liver. Results: The HSP70 protein expression in H0 group was 133%, which was increased compared with that in group C; HSP70 protein expression in H24 group was increased to 175%, which was significantly different from that in group C (P <0.05). The expression of HSP70 protein in liver of E0, E24 and E48 groups was significantly higher than that of C group (P <0.05), showing a gradually increasing trend. The HSP70 protein expression in the liver of HE0 group was 138%, which was also significantly higher than that of the C group (P <0.05); however, the HE24 group decreased to 89%, significantly lower than that of the E24 group (P <0.05) 224%, which was significantly higher than that in group C (P <0.05) and higher than that in E48 group (176%). The difference was statistically significant (P <0.05). Conclusion: Preheating treatment significantly increased hepatic HSP70 expression and delayed expression. Preheating resulted in a significant decrease in GSH but with little or no effect on MDA and SOD. Preheating treatment can not alleviate the trend of GSH decreased significantly after eccentric exercise.
