目的研究蛋白TCTP(translational controlled tumorprotein,翻译控制的肿瘤蛋白)在多发性骨髓瘤(Multiple Myeloma,MM)患者中的表达水平差异及其临床意义。方法采用免疫组化SP法,检测TCTP在MM病变细胞及其它细胞中的定位及表达。利用自动图像分析仪定量分析TCTP的相对含量(灰度)。结果基因TPT1编码的蛋白TCTP在上述样本单个核细胞中均有表达,定位于细胞质中;MM单个核细胞质中TCTP的阳性表达率a=83.2%,平均灰度值m=187.3±17.31,显著强于正常人(a=71.1%,m=159.8±19.02)、白血病患者(a=79.3%,m=166.4±19.27)及非肿瘤疾病患者(a=80.6%,m=170.9±18.46)中的表达,P值均﹤0.05。结论蛋白TCTP在MM中高表达,具有相对特异性。可能成为研究MM发病机理和基因治疗的新靶标。
Objective To study the expression differences of TCTP (translational controlled tumor protein) in patients with multiple myeloma (MM) and its clinical significance. Methods Immunohistochemical SP method was used to detect the localization and expression of TCTP in MM lesion cells and other cells. The relative content of TCTP (grayscale) was quantified using an automated image analyzer. Results The TCTP of gene TPT1 was expressed in all of the above samples and localized in the cytoplasm. The positive expression rate of TCTP in MM mononuclear cytoplasm was 83.2% and the average gray value was 187.3 ± 17.31 (A = 71.1%, m = 159.8 ± 19.02), leukemia patients (a = 79.3%, m = 166.4 ± 19.27) and non-neoplastic disease patients (a = 80.6%, m = 170.9 ± 18.46) , All P <0.05. Conclusion The protein TCTP is overexpressed in MM and has relative specificity. May be a new target for studying the pathogenesis and gene therapy of MM.
