为阐明流行性出血热(EHF)灭活疫苗的细胞免疫保护性机制,对地鼠肾细胞培养灭活疫苗免疫小鼠的脾细胞增殖,产生白细胞介素-2(IL-2)和干扰素(IFN)的能力进行了研究。结果表明,疫苗免疫小鼠脾细胞对ConA及EHF病毒抗原的刺激指数均显著高于正常小鼠。两组小鼠脾细胞在ConA刺激下产生IL-2和IFN,的水平无显著差异,但免疫小鼠受EHF病毒抗原刺激后产生的IL-2水平明显高于正常对照组;15只免疫小鼠有9只对病毒抗原的刺激产生IFN反应;免疫小鼠血清特异性抗体阳转率为93.3%。上述结果对于评价EHF灭活疫苗的免疫效果具有重要价值。
To clarify the cellular immunity protective mechanism of the inactivated vaccine against epidemic hemorrhagic fever (EHF), the spleen cells of mice immunized with the inactivated vaccine of mouse kidney cell culture were proliferated to produce interleukin-2 (IL-2) and interferon (IFN) was studied. The results showed that the stimulation index of ConA and EHF virus antigens in spleen cells of vaccinated mice were significantly higher than that of normal mice. The level of IL-2 and IFN produced by spleen cells stimulated by ConA was no significant difference between the two groups, but the level of IL-2 in immune mice stimulated by EHF virus antigen was significantly higher than that of normal control group. Nine mice had IFN response to virus antigen stimulation; the positive rate of serum specific antibody in immunized mice was 93.3%. The above results for the evaluation of the immune effect of EHF inactivated vaccine has important value.
