目的研究1α,25-(OH)2D3对SD大鼠成骨细胞增殖及相关蛋白的表达的影响。方法采用流式细胞术(FCM)检测细胞周期,RT-PCR和Western blotting分别检测细胞G1期调节蛋白细胞周期素D1(cyclin D1)、细胞周期素依赖性激酶4(CDK4)、p21 mRNA和蛋白的表达。结果与0 nmol·L-1组比较,各1α,25-(OH)2D3干预组细胞周期发生改变,G0/G1期细胞数递减,而S+G2/M期细胞比例升高,PI增加,差异显著(P<0.05);且1α,25-(OH)2D3可诱导细胞cyclin D1、CDK4 mRNA及蛋白表达和下调p21(P<0.05)。结论置1α,25-(OH)2D3可上调cyclin D1、CDK4表达和抑制负性调节因子p21,调节相关蛋白表达以促进成骨细胞增殖。
Objective To investigate the effects of 1α, 25- (OH) 2D3 on the proliferation of osteoblasts and the expression of related proteins in SD rats. Results Compared with 0 nmol·L-1 group, the cell cycle changed in each 1α, 25- (OH) 2D3 group, the number of cells in G0 / G1 phase decreased, while the proportion of cells in S + G2 / M phase increased, 1α, 25- (OH) 2D3 could induce the expression of cyclin D1, CDK4 mRNA and protein and down-regulate p21 (P <0.05). CONCLUSION: 1α, 25- (OH) 2D3 up-regulates the expression of cyclin D1 and CDK4 and inhibits the negative regulator p21, and regulates the expression of related proteins to promote the proliferation of osteoblasts.
