目的建立蜣螂有效部位的毛细管电泳(CE)指纹图谱,探讨利用总量统计矩法分析指纹图谱的可行性。方法采用毛细管区带电泳法(CZE),以弹性石英毛细管柱(60 cm×75μm)为分离通道,75 mmol/L硼砂溶液(pH 9.8)作缓冲液,运行电压+20 kV,检测波长200 nm,指纹图谱的评价采用药典委员会相似度分析软件与总量统计矩法。结果建立了蜣螂有效部位的CE指纹图谱,药典委员会的相似度软件与总量统计矩法计算的相似度基本一致,显示蜣螂有效部位批间差异小。结论建立的CE指纹图谱准确简便、重现性好,可作为蜣螂提取物的质量控制方法,而总量统计矩法可以作为指纹图谱的分析方法。
Objective To establish capillary electrophoresis (CE) fingerprinting of effective parts of Dung beetle, and to explore the feasibility of using total statistical moment method to analyze fingerprints. Methods The capillary zone electrophoresis (CZE) method was used. The column was equipped with a flexible quartz capillary column (60 cm × 75 μm) as the separation channel and 75 mmol / L borax solution (pH 9.8) The fingerprints were evaluated by Pharmacopoeia Commission similarity analysis software and total statistical moment method. Results The CE fingerprint of the effective parts of Dung beetle was established. The similarities between the similarity software of the Pharmacopoeia Commission and the total statistical moment method were basically the same, which showed that the differences between the effective parts of Dung beetle were small. Conclusion The established CE fingerprint is accurate and reproducible, and can be used as a quality control method for the extract of dung beetle. The total statistical moment method can be used as an analytical method for fingerprint analysis.
