目的:观察温补肾阳汤对哮喘小鼠外周血、肺组织Eotaxin表达的影响,探讨其抑制气道炎症作用。方法:健康雌性BALB/c小鼠50只,随机分为空白对照组、模型组、醋酸泼尼松组、温补肾阳汤组、温补肾阳汤+醋酸泼尼松组,每组10只。卵白蛋白(OVA)致敏及激发造模方法建立动物模型。计数肺泡灌洗液EOS;光镜下观察肺组织病理学改变;ELISA检测外周血Eotaxin表达;免疫组织化学检测肺组织Eotaxin蛋白水平。结果:药物干预组小鼠外周血、肺组织Eotaxin表达较模型组明显降低(P<0.01),肺泡灌洗液EOS计数明显减少(P<0.01),其中温补肾阳汤+醋酸泼尼松组较温补肾阳汤组外周血、肺组织Eotaxin表达及肺泡灌洗液EOS计数均降低(P<0.05),但与醋酸泼尼松组无差异(P>0.05)。结论:温补肾阳汤可能通过降低哮喘小鼠体内Eotaxin水平,减轻炎症细胞在肺组织局部浸润,从而抑制气道炎症。温补肾阳汤与醋酸泼尼松有一定协同作用,可减轻或避免大剂量糖皮质激素引起的不良反应。
Objective: To observe the effects of Warming Kidney Decoction on the expression of Eotaxin in peripheral blood and lung tissue of asthmatic mice, and to explore its inhibitory effect on airway inflammation. Methods: Fifty healthy female BALB / c mice were randomly divided into blank control group, model group, prednisone acetate group, warm-tonifying kidney-yang decoction group, warming-tonifying kidney-yang decoction + prednisone acetate group. Ovalbumin (OVA) sensitization and stimulation modeling methods to establish animal models. EOS of bronchoalveolar lavage fluid was counted. Pathological changes of lung tissue were observed under light microscope. Eotaxin expression in peripheral blood was detected by ELISA. Eotaxin protein expression in lung tissue was detected by immunohistochemistry. Results: The expression of Eotaxin in the peripheral blood and lung tissue of the drug-treated mice was significantly lower than that of the model group (P <0.01), and the count of EOS in the bronchoalveolar lavage fluid was significantly decreased (P <0.01). The Wenshen Shenyang decoction + prednisone acetate group The expression of Eotaxin in peripheral blood and lung tissue and the count of EOS in bronchoalveolar lavage fluid in the Warming Kidney-Yang Decoction group were all decreased (P <0.05), but not in the prednisone acetate group (P> 0.05). Conclusion: Warming Kidney Yang Decoction may inhibit airway inflammation by decreasing Eotaxin level in asthmatic mice and decreasing local infiltration of inflammatory cells in lung tissue. Warming Kidney Soup and prednisone acetate have a synergistic effect, can reduce or avoid the adverse reactions caused by high-dose glucocorticoid.
