目的:观察四物汤对血虚证小鼠肝细胞凋亡及促凋亡基因Bax表达的影响。方法:30只C57BL/6J小鼠随机分为正常组、模型组、四物汤组。以腹腔注射环磷酰胺建立血虚证模型,采用TUNEL法检测肝细胞凋亡,荧光定量PCR法检测Bax mRNA表达。结果:四物汤可升高血虚证模型小鼠外周血红细胞计数、白细胞计数以及血红蛋白含量,降低肝细胞凋亡百分率,下调促凋亡基因Bax mRNA表达。结论:四物汤补益肝血作用机理可能与其减少肝细胞凋亡有关。
Objective: To observe the effect of Si Wu Tang on hepatocyte apoptosis and the expression of Bax gene in mice with blood deficiency syndrome. Methods: Thirty C57BL / 6J mice were randomly divided into normal group, model group and Siwu Tang group. The model of blood deficiency was established by intraperitoneal injection of cyclophosphamide. TUNEL method was used to detect the apoptosis of hepatocytes. The expression of Bax mRNA was detected by quantitative PCR. Results: Siwu Decoction could increase peripheral blood erythrocyte count, white blood cell count and hemoglobin, decrease the percentage of hepatocyte apoptosis, and down-regulate the expression of pro-apoptotic gene Bax mRNA in mice with blood deficiency syndrome. Conclusion: The mechanism of Treating Liver-Blood with Si-Wu-Tang may be related to reducing hepatocyte apoptosis.
