目的:检测HAI-1mRNA和ST14 mRNA在非小细胞肺癌组织与正常肺组织中的表达,并分析其与临床病理的关系。方法:应用原位杂交技术(In Situ Hybridization),积分光密度半定量,检测ST14mRNA和HAI-1mRNA在非小细胞肺癌及相应的正常肺组织中的表达。结果:HAJ-1mRNA和ST14mRNA在非小细胞肺癌组织中表达低与正常肺组织(p<0.05),在高分化癌组织表达高于低分化组织(p<0.05),在无淋巴结转移组表达高于有淋巴结转移组(p<0.05),与年龄,性别,癌组织类型无关(p>0.05)。结论:HAI-1mRNA与ST14mRNA的表达在肺癌与正常组织中存在差异,在正常组织中表达高于肺癌组织。提示在肿瘤进程中,HAI-1mRNA和ST14mRNA转录受抑制,二基因在肺癌中可能表现为抑癌基因。HAI-1mRNA和ST14mRNA与肺癌的分化程度有关,在高分化的癌组织中表达高于低分化癌组织,在有淋巴转移组低于无淋巴转移组,提示此2基因可能参与促进细胞分化成熟,并且其表达降低可能促进肿瘤的转移。
OBJECTIVE: To detect the expression of HAI-1 mRNA and ST14 mRNA in non-small cell lung cancer and normal lung tissue, and to analyze their relationship with clinical pathology. Methods: The expression of ST14 mRNA and HAI-1 mRNA in non-small cell lung cancer and corresponding normal lung tissues were detected by In Situ Hybridization and semi-quantitative method of integral optical density. Results: The expression of HAJ-1 mRNA and ST14 mRNA in non-small cell lung cancer tissues was significantly lower than that in poorly differentiated tissues (p <0.05), but higher in non-small cell lung cancer tissues (p <0.05) There was no correlation between lymph node metastasis (p <0.05) and age, sex, type of cancer (p> 0.05). Conclusion: The expression of HAI-1mRNA and ST14mRNA in lung cancer and normal tissues are different, and they are higher in normal tissues than in lung cancer tissues. Tip in the tumor progression, HAI-1mRNA and ST14mRNA transcription is inhibited, two genes may appear in lung cancer tumor suppressor gene. The expression of HAI-1mRNA and ST14mRNA correlated with the degree of lung cancer differentiation. The expression of HAI-1mRNA and ST14mRNA in well-differentiated cancers was higher than that in poorly differentiated cancers. The expression of HAI-1mRNA and ST14mRNA in lymphatic metastasis group was lower than that in lymphatic metastasis group, suggesting that these two genes may be involved in promoting cell differentiation and maturation, And its decreased expression may promote tumor metastasis.
