目的:应用经皮腔内冠状动脉成形术球囊堵闭猪冠状动脉建立急性心肌梗死动物模型,并通过TTC染色识别急性心肌梗死动物模型中存活心肌和梗死心肌。方法:普通家猪13只,麻醉气管插管后行冠状动脉造影术,置入经皮腔内冠状动脉成形术球囊至冠状动脉左前降支远端,堵闭血流60~90 min,建立家猪心肌梗死模型。处死试验动物取出心脏,将心肌组织切成薄片后采用TTC染色法识别存活心肌。结果:家猪均完成冠状动脉左前降支远端的封堵,2只家猪分别在堵闭45 min,65 min时因心室颤动死亡,存活的11只家猪均成功建立急性心肌梗死模型,TTC染色显示正常心肌88个节段,梗死心肌49个节段,存活心肌39个节段。结论:运用经皮腔内冠状动脉成形术球囊封堵冠状动脉可成功建立猪急性心肌梗死模型,通过TTC染色能识别出心肌梗死模型中的梗死心肌与存活心肌。
OBJECTIVE: To establish an animal model of acute myocardial infarction (AMI) by percutaneous transluminal coronary angioplasty balloon occlusion and to identify viable myocardium and infarct myocardium in animal models of acute myocardial infarction by TTC staining. Methods: Thirteen ordinary domestic pigs were anesthetized with tracheal intubation and underwent coronary angiography. The percutaneous transluminal coronary angioplasty balloon was inserted into the distal left anterior descending coronary artery to occlude the blood flow for 60-90 min to establish Domestic pig myocardial infarction model. Animals were sacrificed to remove the heart, the myocardial tissue was cut into thin slices using TTC staining to identify viable myocardium. Results: The domestic pigs completed the occlusion of the distal left anterior descending coronary artery. Two domestic pigs died of ventricular fibrillation at 45 min and 65 min after occlusion, respectively. All the 11 surviving pigs successfully established acute myocardial infarction model, TTC staining showed 88 segments of normal myocardium, 49 segments of infarcted myocardium and 39 segments of viable myocardium. Conclusion: Percutaneous transluminal coronary angioplasty with balloon occlusion of coronary arteries can successfully establish a model of acute myocardial infarction in pigs, and myocardial infarction and viable myocardium can be identified by TTC staining.
