目的 :探讨诱生型一氧化氮合酶 (iNOS)在喉癌中的表达及其临床意义。方法 :采用逆转录 -聚合酶链反应 (RT -PCR)技术检测 40例不同病理分化程度、临床分期的喉鳞癌组织、癌旁组织及 1 0例喉乳头状瘤、7例正常喉黏膜中诱生型一氧化氮合酶 (iNOS)mRNA的表达。结果 :喉癌组织iNOSmRNA阳性表达率为 85.0 0 % ,较对照组癌旁组织 ( 2 0 .0 0 % )、良性肿瘤组 ( 4 0 .0 0 % )、正常人组 ( 0 7)明显增高 (P <0 .0 0 0 1 ,P <0 .0 0 5,P <0 .0 0 0 1 ) ,有淋巴结转移组 ( 1 9 1 9)显著高于无淋巴结转移组 ( 71 .42 % ) (P <0 .0 5) ,iNOSmRNA阳性表达与肿瘤原发部位及T分级无关 ,与细胞分化程度呈负相关。结论 :喉癌组织中存在iNOS ,它可能通过合成一氧化氮 (NO)在分子水平参与了喉癌的发生、发展。
Objective: To investigate the expression of inducible nitric oxide synthase (iNOS) in laryngeal carcinoma and its clinical significance. Methods: Forty cases of laryngeal squamous cell carcinoma, paracancer tissues and 10 cases of laryngeal papilloma of different pathological differentiation degree and clinical stages were detected by reverse transcription-polymerase chain reaction (RT-PCR) Expression of inducible nitric oxide synthase (iNOS) mRNA. Results: The positive expression rate of iNOS mRNA in laryngeal carcinoma was 85.0%, which was significantly higher than that in the control group (20.0%), benign tumor group (40.0%) and normal human group (0 7) (P <0.001, P <0.05, P <0.001), with lymph node metastasis (1 9 1 9) was significantly higher than those without lymph node metastasis (71.42% ) (P <0.05). The positive expression of iNOS mRNA was not related to the primary tumor site and T grade, but negatively correlated with the degree of cell differentiation. CONCLUSION: iNOS is present in laryngeal carcinoma, which may be involved in the development of laryngeal carcinoma at the molecular level through the synthesis of nitric oxide (NO).
