细胞激光扫描断层(CT)技术系借助于现代先进的激光扫描共焦聚显微镜(CLSM)来实现的。研究了本室系列抗癌中药之一的抗癌宝Ⅱ号对小鼠腹水癌细胞的作用。以细胞CT、光切0.6μ、三维重组、立体旋转和DNA荧光强度分布图像进行分析,结果发现抗癌宝Ⅱ号使癌细胞核DNA荧光物质凝聚、裂解,分布图呈切齿状,荧光强度明显减弱,与未用药的对照组有明显区别;并发现分裂期的染色体碎裂。这种关于单细胞CT技术,能达到荧光显微镜、流式细胞仪、电子显微镜所达不到的效果,因此使中药抗癌机理的研究提高到一个新的水平。
The Cellular Laser Scanning Tomography (CT) technology is implemented using a modern advanced laser scanning confocal polymicroscope (CLSM). The effects of anti-cancer Baohao No. II, one of the series of anti-cancer herbs in this family, on mouse ascites cancer cells were studied. The analysis of cell CT, light-cutting 0.6μ, three-dimensional recombination, three-dimensional rotation, and DNA fluorescence intensity distribution image showed that anti-cancer treasure II condenses and cleaves the nuclear DNA fluorescent material of cancer cells, and the distribution diagram is dentate, and the fluorescence intensity is obvious. Weakened, with a clear difference between the untreated control group; and found that the disintegrating chromosome fragmented. This kind of single-cell CT technology can achieve the effects that fluorescence microscope, flow cytometry and electron microscope can not reach, thus raising the research of Chinese traditional medicine anti-cancer mechanism to a new level.
