目的:探讨中药复方消斑通脉合剂药物血清对血小板源性生长因子(PDGF-BB)诱导的大鼠血管平滑肌细胞(VSMC)增殖与细胞周期分布的影响及相关作用机制。方法:体外培养VSMCs,PDGF-BB刺激VSMCs,采用MTT法检测药物血清对VSMCs增殖的影响;流式细胞仪分析各组细胞周期分布情况;应用Western印迹法检测药物血清对VSMCs中P21waf1/cip1蛋白的表达变化。结果:MTT法显示药物血清呈剂量依赖性抑制VSMCs增值,高剂量与低剂量比较有显著差异(P(0.05);流式细胞仪分析结果显示与模型组比较,低剂量与高剂量组明显提高G0/G1期百分率(P(0.01)、减少S期百分率(P(0.01),低剂量与高剂量组比较有显著差异(P(0.05);Western印迹法显示与模型组比较,高剂量与低剂量可明显提高VSMCs中P21waf1/cip1蛋白表达(P(0.01),低剂量与高剂量比较有统计学差异(P(0.01)。结论:中药复方消斑通脉合剂药物血清通过促进VSMCs中P21waf1/cip1蛋白表达,阻滞VSMCs细胞周期进程,起到抑制VSMCs增殖作用。
Objective: To investigate the effects of Xiaobantongmai Mixture of traditional Chinese medicine on proliferation and cell cycle distribution of rat vascular smooth muscle cells (VSMCs) induced by platelet-derived growth factor (PDGF-BB) and its related mechanism. Methods: VSMCs were cultured in vitro and PDGF-BB was used to stimulate VSMCs. The effect of serum on the proliferation of VSMCs was detected by MTT assay. The distribution of cell cycle was analyzed by flow cytometry. Western blotting was used to detect the expression of P21waf1 / cip1 protein in VSMCs Changes in expression. Results: The MTT assay showed that the drug serum increased the proliferation of VSMCs in a dose-dependent manner. There was significant difference between the high dose and the low dose (P <0.05). Flow cytometry analysis showed that compared with the model group, the low dose and high dose groups significantly increased Compared with the model group, the percentage of G0 / G1 phase (P (0.01), the percentage of S phase decreased (P (0.01)), the low dose and the high dose group were significantly different P21waf1 / cip1 protein expression was significantly increased in VSMCs (P <0.01), and there was a significant difference between low dose and high dose (P (0.01) .Conclusion: Traditional Chinese medicine Xiaobantongmai Mixture can improve serum P21waf1 / cip1 protein expression, blocking VSMCs cell cycle progression, play a role in inhibiting the proliferation of VSMCs.
