目的检测人肾透明细胞癌组织中细胞凋亡相关重要因子Caspase-3、Caspase-9和增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)表达的变化,研究这些因子的表达与肾癌的关系。方法标本离体后分成3组,①肾癌细胞组;②癌旁组织组;③正常肾组织组。均用10%福尔马林固定48h,常规石蜡包埋、切片5μm,应用免疫组织化学方法进行观察。结果 PCNA免疫组织化学染色结果,正常肾组织内的细胞核有弱的阳性表达,癌旁组织阳性表达增加,高于正常肾组织;肾癌组织阳性表达明显增强,明显高于癌旁组织。Caspase-3与Caspase-9均在正常肾组织中有弱的阳性反应,癌旁组织阳性表达增加,肾癌组织表达明显减少。3组之间两两比较差异均有统计学意义(P<0.01)。肾癌4个期之间比较差异无统计学意义(P>0.05)。结论癌旁组织Caspase-3和Caspase-9蛋白表达明显高于正常肾组织及癌组织,癌组织表达最少;癌组织中Caspase-3和Caspase-9表达下降,说明细胞凋亡减少;PCNA在正常肾组织组、癌旁组织组、肾癌组织组的表达呈明显加强趋势,癌组织细胞增殖旺盛,是肿瘤形成的重要机制。Caspase-3、Caspase-9和PCNA于肾透明细胞癌的表达在肿瘤分期间差异无统计学意义。Caspase-3与Caspase-9蛋白表达呈正相关。
Objective To detect the expression of Caspase-3, Caspase-9 and proliferating cell nuclear antigen (PCNA) in human renal clear cell carcinoma tissues and to study the relationship between the expression of these factors and renal cell carcinoma. Methods The specimens were divided into 3 groups in vitro, ① kidney cancer cell group; ② paracancer tissue group; ③ normal kidney tissue group. All were fixed with 10% formalin for 48h, embedded in paraffin and sliced 5μm, and observed by immunohistochemical method. Results The results of PCNA immunohistochemical staining showed that the nucleus in normal renal tissue was weakly positive and the expression in paracancerous tissues was higher than that in normal renal tissues. The positive expression of PCNA in renal cell carcinoma was significantly higher than that in paracancerous tissues. Caspase-3 and Caspase-9 in normal renal tissue have a weak positive reaction, paracancer tissue positive expression increased significantly reduced expression of renal cell carcinoma. The differences between the three groups were statistically significant (P <0.01). There was no significant difference between the 4 stages of renal cell carcinoma (P> 0.05). Conclusions The expressions of Caspase-3 and Caspase-9 in paracancerous tissues are significantly higher than those in normal renal tissues and cancerous tissues, with the least in cancerous tissues. The decreased expressions of Caspase-3 and Caspase-9 in cancerous tissues suggest that the apoptosis of PCNA decreases The expression of renal tissue, paracancer and renal cell carcinoma tissues showed an obvious trend of enhancement. The proliferation of cancer cells was an important mechanism of tumor formation. There was no significant difference in the expression of Caspase-3, Caspase-9 and PCNA in renal clear cell carcinoma during the tumor stage. Caspase-3 and Caspase-9 protein expression was positively correlated.
