目的:观察蠲哮汤对哮喘大鼠模型气道重塑细胞增殖信号通路的影响。方法:60只SPF级雌性SD大鼠,随机分为6组:正常组,模型组,地塞米松组,蠲哮汤低、中、高剂量组,每组10只。除正常组外,各组均采用卵蛋白(OVA)联合雾化吸入的方法建立哮喘大鼠模型。采用实时荧光定量PCR检测各组大鼠肺组织中细胞外信号调节激酶(ERK)1、ERK2及磷脂酰肌醇3激酶(PI3K)mRNA的表达水平。结果:与正常组比较,模型组ERK1、ERK2和PI3K mRNA表达上调(P<0.05);与模型组比较,各治疗组大鼠ERK1、ERK2和PI3K mRNA表达下调(P<0.05)。结论:蠲哮汤可以有效抑制哮喘大鼠的ERK1、ERK2和PI3K mRNA表达,达到调控细胞增殖信号通路的目的,从而抑制哮喘大鼠气道重塑的进程。
Objective: To observe the effect of “Xue-Hua Decoction” on proliferation signal pathway of airway remodeling in asthmatic rat model. Methods: Sixty SPF female SD rats were randomly divided into 6 groups: normal group, model group, dexamethasone group and low dose asthma group, 10 rats in each group. Except the normal group, the asthma rat model was established by ovalbumin (OVA) combined with atomization inhalation. Real-time fluorescence quantitative PCR was used to detect the expression of extracellular signal-regulated kinase (ERK) 1, ERK2 and phosphoinositide 3-kinase (PI3K) mRNA in the lung tissue of each group. Results: Compared with normal group, ERK1, ERK2 and PI3K mRNA expressions were up-regulated in model group (P <0.05). Compared with model group, the expression of ERK1, ERK2 and PI3K mRNA were down-regulated in all treatment groups. Conclusion: Astragalus soup can effectively inhibit the expression of ERK1, ERK2 and PI3K mRNA in asthmatic rats and regulate the cell proliferation signal pathways, thereby inhibiting the process of airway remodeling in asthmatic rats.
