目的 探讨良性前列腺增生症 (BPH)各区细胞增殖和凋亡特征及其与BPH发病的关系。 方法 采用流式细胞术对 33例BPH病人前列腺各区细胞表皮生长因子 (EGF)、表皮生长因子受体 (EGF R)、碱性纤维细胞生长因子 (bFGF)和转化生长因子 β1(TGF β1)表达、细胞DNA含量和细胞凋亡情况进行定量对比分析。 结果 EGF和EGF R的标记率和表达量在移行区 (TZ)明显高于周边区 (PZ)和中央区 (CZ) (P均 <0 .0 1)。bFGF和TGF β1的标记率在三个区差异无显著性 (P>0 .0 5 ) ,而bFGF和TGF β1的表达量在TZ明显高于PZ和CZ(P均 <0 .0 1)。TZ细胞增殖指数(PI)明显高于PZ和CZ(P <0 .0 1) ;各区细胞凋亡率差异无显著性。 结论 TZ细胞中EGF、EGF R、bFGF和TGF β1高表达和高增殖活性是BPH发病的重要因素 ,细胞凋亡与BPH发生特征无明显相关。
Objective To investigate the characteristics of cell proliferation and apoptosis in benign prostatic hyperplasia (BPH) and its relationship with the pathogenesis of BPH. Methods Flow cytometry was used to detect the expression of epidermal growth factor (EGF), basic fibroblast growth factor (bFGF) and transforming growth factor β1 (TGFβ1) in the prostate of 33 BPH patients. , DNA content and apoptosis were quantitatively analyzed. Results The labeling rates and expression levels of EGF and EGF R in TZ were significantly higher than those in peripheral zone (PZ) and central zone (CZ) (P <0.01). There was no significant difference in the labeling rates of bFGF and TGFβ1 among the three regions (P> 0.05), while the expression of bFGF and TGFβ1 in TZ was significantly higher than that in PZ and CZ (P <0.01). TZ cell proliferation index (PI) was significantly higher than that of PZ and CZ (P <0.01). There was no significant difference in apoptosis rate among different regions. Conclusion The high expression and high proliferative activity of EGF, EGFR, bFGF and TGFβ1 in TZ cells is an important factor in the pathogenesis of BPH. There is no significant correlation between the apoptosis and the occurrence of BPH.
