为研究急性呼吸窘迫综合征血管内皮细胞功能改变,采用同种骨髓提取液(BME)复制兔急性呼吸窘迫综合征(ARDS)模型,连续多时相点检测血管内皮标记物:循环内皮细胞(CEC)、血浆颗粒膜蛋白-140(GMP-140)和内皮素(ET-1)以及肺组织GMP-140含量,并行常规病理及电镜检查。发现致伤后0.5h,CEC、血浆GMP-140、ET-1 均显著升高,持续6h 仍显著高于伤前和对照,其中血浆GMP-140 升高达3.25 倍,1h 时达到高峰,而肺组织GMP-140 含量降低。血浆GMP-140 早期升高,幅度较大,与PaO2 负相关,与其它指标相关性亦较好。提示:经兔颈静脉恒流速注射BME(0.35m l/kg,2m l/h)能复制出理想的ARDS模型。检测血浆GMP-140可能是临床ARDS早期诊断、病情监测和评估的一个重要手段
In order to study the functional changes of vascular endothelial cells in acute respiratory distress syndrome (ARR), allogeneic bone marrow extract (BME) was used to replicate acute respiratory distress syndrome (ARDS) model in rabbits and the vascular endothelial markers (CECs) , Plasma membrane protein-140 (GMP-140) and endothelin (ET-1) and lung tissue GMP-140 content, routine pathology and electron microscopy. The levels of CEC, plasma GMP-140 and ET-1 were significantly increased at 0.5 h after injury, and remained significantly higher at 6 h than pre-injury and control, with plasma GMP-140 up 3.25 fold and peaked at 1 h , While the lung tissue GMP-140 content decreased. Plasma GMP-140 increased early, with a greater magnitude, negative correlation with PaO2, and other indicators of the correlation is also good. It is suggested that the ideal ARDS model can be replicated by injecting BME (0.35m l / kg, 2ml / h) at a constant flow rate in rabbit jugular vein. Detecting plasma GMP-140 may be an important means of early diagnosis, condition monitoring and assessment of clinical ARDS
