目的 探讨肿瘤坏死因子 - α(TNF- α)在局灶脑缺血损伤中神经元的表达机制。方法 本研究应用逆转录一多聚酶链反应 (RT- PCR)方法检测大鼠局灶脑缺血模型中 TNF- αm RNA的表达。结果 实验发现缺血以皮层为主的 TNF- αm RNA表达随时间变化。缺血 1h即可观察到 TNF- αm RNA的表达 ,3h至 5 d明显增加 ,12 h表达达高峰。我们还观察到缺血区中心以底节为主的白质神经纤维、纹状体及海马中存在轻度不同 TNF-αm RNA的表达。对照组及缺血对侧均未见到此表达。结论 脑缺血后 TNF-α增加 ,是加重缺血性脑损伤的重要因素 ,拮抗TNF- α可能是治疗脑梗死的一条途径。
Objective To investigate the expression of tumor necrosis factor-α (TNF-α) in neurons during focal cerebral ischemia. Methods In this study, reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of TNF-αmRNA in focal cerebral ischemia model of rats. Results The experiment found that the cortical TNF-αm RNA expression changes with time. TNF-αmRNA expression was observed at 1h after ischemia, increased significantly from 3h to 5d, and peaked at 12h. We also observed the presence of mildly different TNF-αmRNA expression in white matter nerve fibers, striatum and hippocampus in the basal ganglia in the ischemic region. The control group and the opposite side of ischemia did not see this expression. Conclusions Increased TNF-α after cerebral ischemia is an important factor in aggravating ischemic brain injury. Antagonizing TNF-α may be a way to treat cerebral infarction.
