通过根尖染色体鉴定、田间农艺性状观测、桔梗皂苷-D含量测定和比较、蛋白质含量测定和蛋白质聚丙烯酰胺凝胶电泳实验,对桔梗同源四倍体进行了遗传稳定性研究。结果表明:桔梗四倍体株系的染色体数目仍为36条,农艺性状保持了典型的多倍体特征,桔梗皂苷-D含量及蛋白质含量均高于对照二倍体株系,各株系蛋白质电泳谱带基本相似。实验结果说明桔梗同源四倍体具有较好的遗传稳定性,为药用植物多倍体育种技术的发展和生产应用提供了依据。
The genetic stability of autotetraploid of Campanulaceae was studied by apical chromosome identification, field agronomic traits, determination and comparison of platycodin-D content, protein content determination and protein polyacrylamide gel electrophoresis. The results showed that the number of chromosomes in tetraploid lines of Platycodon grandiflorum was still 36, and the agronomic traits maintained typical polyploid characters. The platycodin-D content and protein content of platycodon were higher than that of the control diploid lines. The protein content Electrophoresis bands are similar. The experimental results showed that the autotetraploid of Campanulaceae has good genetic stability and provided the basis for the development, production and application of the polyploid breeding technology of medicinal plants.
