目的研究参葛降糖胶囊对胰岛素抵抗糖尿病大鼠脂肪细胞因子水平的影响,探讨参葛降糖胶囊治疗糖尿病的作用机制。方法采用灌胃高脂高糖脂肪乳加腹腔注射小剂量链脲佐菌素(STZ)的方法建立胰岛素抵抗糖尿病大鼠模型。造模成功后随机分为正常对照组、模型组、阳性对照组和参葛降糖胶囊低、中、高剂量组,分别给予相应的干预,共30天。检测各组大鼠总胆固醇(TC)、三酰甘油(TG)、游离脂肪酸(FFA)、瘦素(leptin)和肿瘤坏死因子-α(TNF-α)水平。结果与正常对照组比较,参葛降糖胶囊中、高剂量组TC、TG、TNF-α、leptin含量差异无统计学意义(P>0.05),而FFA含量差异有统计学意义(P<0.001,P<0.01);参葛降糖胶囊低剂量组大鼠血清TC、TG、TNF-α、FFA含量差异有统计学意义(P<0.001,P<0.01,P<0.05),而leptin水平差异无统计学意义(P>0.05)。与模型组比较,参葛降糖胶囊高、中剂量组大鼠血清FFA、TC、TG、leptin、TNF-α含量降低显著,差异有统计学意义(P<0.001,P<0.01);参葛降糖胶囊低剂量组大鼠血清TC、TG、leptin含量降低,差异有统计学意义(P<0.001,P<0.01,P<0.05),而FFA、TNF-α含量差异无统计学意义(P>0.05)。结论参葛降糖胶囊可以通过影响胰岛素抵抗糖尿病大鼠脂肪细胞因子水平,调节脂代谢,改善胰岛素抵抗。
Objective To study the effects of gongjiang jiangtang capsule on adipokines in diabetic rats with insulin resistance and to explore the mechanism of gingjiang tangtang capsule in treating diabetes mellitus. Methods The model of insulin-resistant diabetic rats was established by intragastric administration of high-fat, high-fat and fat-free fat emulsion and intraperitoneal injection of low dose streptozotocin (STZ). After successful modeling, the rats were randomly divided into normal control group, model group, positive control group and low, medium and high dosage of ginseng hypoglycemic capsules, and the corresponding interventions were given for 30 days respectively. The levels of total cholesterol (TC), triglyceride (TG), free fatty acid (FFA), leptin and tumor necrosis factor-α (TNF-α) in rats were measured. Results Compared with the normal control group, the contents of TC, TG, TNF-α and leptin in Shenkeliangtang capsule group were not significantly different (P> 0.05), but there were significant differences in the content of FFA (P <0.01, P <0.01). The contents of TC, TG, TNF-α and FFA in the low-dose Shengeng Jiangtang capsule group were significantly different (P <0.001, P < No statistical significance (P> 0.05). Compared with model group, the contents of serum FFA, TC, TG, leptin and TNF-α of Shengeng Jiangtang capsule high and middle dose groups decreased significantly (P <0.001, P <0.01) The levels of TC, TG and leptin in the low-dose Jiangtang capsule low-dose group decreased significantly (P <0.001, P <0.01, P <0.05), while there were no significant differences in the levels of FFA and TNF- > 0.05). Conclusion GSP capsule can affect adiponectin level in insulin-resistant diabetic rats, regulate lipid metabolism and improve insulin resistance.
