[目的]研究电子线路板粉末喂饲雄性小鼠,对其脏器尤其是生殖系统的毒性作用。[方法]雄性小鼠分别喂饲常规饲料和含电子线路板粉末的混合饲料,定期记录体质量和摄食量,90d后处死,取脑、心、肝、脾、肺、肾和睾丸称重并进行病理切片观察,计算脏器系数,测定肝、脑中多溴联苯醚(PBDEs)的含量,测定精子活力及相关动力学参数,免疫荧光法观察睾丸组织Connexin43蛋白表达。[结果]电子线路板粉末24h浸出液致小鼠急性半数致死量(LD50)大于10000mg/kg。经过90d亚慢性实验,实验组小鼠主要脏器系数[肝(4.63±0.39)、肾(1.72±0.29)、脑(1.02±0.13)、肺(0.51±0.04)]比对照组相应脏器系数[肝(3.99±0.42)、肾(1.38±0.16)、脑(0.85±0.15)、肺(0.46±0.06)]明显升高,并且肝、肾出现病理损伤。实验组肝、脑中PBDEs含量分别为175.54ng/g湿重、29.60ng/g湿重,较对照组肝(2.16ng/g湿重)、脑(0.12ng/g湿重)大幅上升。实验组和对照组相比,睾丸的脏器系数、病理切片和Connexin43蛋白表达均未出现明显改变,精子活力及相关动力学参数差异也未见统计学意义。[结论]成年雄性小鼠喂饲电子线路板粉末90d后,肝脏和肾脏出现明显病理学改变,生殖系统未观察到毒性效应。
[Objective] To study the toxic effects of electronic circuit board powder feeding male mice on the organs, especially the reproductive system. [Methods] Male mice were fed with a mixture of conventional diet and electronic circuit board powder. The body weight and food intake were recorded regularly. After 90 days, the mice were sacrificed and the brain, heart, liver, spleen, lung, kidney and testis were weighed The pathological sections were observed and the organ coefficients were calculated. The levels of polybrominated diphenyl ethers (PBDEs) in the liver and brain were measured. The sperm motility and related kinetic parameters were determined. The expression of connexin43 in testis was observed by immunofluorescence. [Results] The acute half-lethal dose (LD50) of mice induced by electronic circuit board powder 24 h was more than 10000 mg / kg. After 90 days of subchronic experiment, the main organ coefficients [liver (4.63 ± 0.39), kidney (1.72 ± 0.29), brain (1.02 ± 0.13), lung (0.51 ± 0.04) The liver (3.99 ± 0.42), kidney (1.38 ± 0.16), brain (0.85 ± 0.15) and lung (0.46 ± 0.06) were significantly increased, and pathological injury occurred in liver and kidney. The content of PBDEs in the liver and brain of the experimental group were 175.54ng / g wet weight and 29.60ng / g wet weight respectively, which were significantly higher than those of the control group (2.16ng / g wet weight) and brain (0.12ng / g wet weight). Compared with the control group, there was no significant change in the organ coefficient, pathological section and Connexin43 protein in the test group and the sperm motility and related kinetic parameters. [Conclusion] The pathological changes of liver and kidney were observed in adult male mice after 90 days feeding of the electronic circuit board powder, and no toxic effects were observed in the reproductive system.
