目的探讨芪珀生脉颗粒对心气虚模型小鼠心肌结构、超氧化物歧化酶(SOD)、丙二醛(MDA)及血清肿瘤坏死因子α(TNF-α)的影响。方法昆明种小鼠84只,随机选取14只(雌雄各半)作为正常组,其余为造模组70只。造模组小鼠每天给予强迫负重游泳至力竭,实验全过程控食,第21天起每天灌服大剂量心得安溶液,连续4天,制造小鼠心气虚模型。实验第11天,将造模组随机分为模型组、补心气组及芪珀生脉颗粒大、中、小剂量组,分别灌胃给药。实验第25天取材,透射电镜观察心肌超微结构的变化,心肌匀浆测定SOD、MDA,眼球取血测血清TNF-α。结果透射电镜下可见,与正常组比较,模型组肌原纤维排列紊乱,线粒体堆积,嵴溶解;各给药组心肌结构损伤均有减轻。与正常组相比,模型组心肌SOD活力显著降低,心肌MDA含量显著升高,血清TNF-α含量显著升高,差异均有统计学意义(P<0.01);与模型组相比,芪珀生脉颗粒大剂量组心肌SOD活力显著升高,心肌MDA含量显著降低,血清TNF-α含量显著降低(P<0.01),差异均有统计学意义(P<0.01)。结论芪珀生脉颗粒能显著提高心肌SOD活力,降低心肌MDA含量,减少血清TNF-α含量,提高机体抗自由基损害的能力,抑制炎症因子对细胞的损害,从而达到保护心肌的作用。
Objective To investigate the effects of Qipeng Shengmai Granule on myocardial structure, superoxide dismutase (SOD), malondialdehyde (MDA) and serum tumor necrosis factor-α (TNF-α) in mice with cardiac Qi deficiency. Methods Kunming mice 84, 14 randomly selected (male and female) as the normal group, the rest of the model group of 70. The mice in model group were forced to swim to exhaustion every day, and the whole experiment was controlled by food. From the 21st day, the large dose of propranolol solution was instilled daily for 4 consecutive days. On the 11th day of the experiment, the model group was randomly divided into model group, Bu Xinqi group and Qibei Shengmai Granule large, medium and small dose groups, respectively, by intragastric administration. On the 25th day of experiment, the changes of myocardial ultrastructure were observed by transmission electron microscope, SOD and MDA were measured in myocardial homogenate and TNF-α in serum by eyeball. Results The results of TEM showed that the myofibrils in the model group were disordered, the mitochondria were accumulated and the crest was dissolved. Compared with the normal group, myocardial injury in each group was alleviated. Compared with the normal group, the activity of SOD in the model group was significantly lower, the content of MDA in the myocardium was significantly increased and the level of TNF-α in the serum was significantly increased (P <0.01). Compared with the model group, Compared with the control group, the activity of SOD in myocardium was significantly increased in the high-dose Shengmai Granules group, the content of MDA in myocardium was significantly decreased and the level of TNF-α in serum was significantly decreased (P <0.01). CONCLUSION: Qibei Shengmai Granule can significantly increase the activity of SOD in myocardium, decrease the content of MDA in myocardium, decrease the content of TNF-α in serum, improve the ability of anti-free radical damage, and inhibit the damage of inflammatory cytokines, so as to protect myocardium.
