目的 观察大鼠三叉神经中脑核 (Vme)内谷氨酸脱羧酶 (GAD)样阳性终末与磷酸激活的谷氨酰胺酶(PAG)、GABAA 受体α3亚单位 (GABAARα3)和小白蛋白 (PV)样阳性神经元之间的联系。 方法 免疫荧光组织化学三重染色技术 ,在激光共聚焦显微镜下进行观察。 结果 在Vme内可观察到大量的GABAARα3样、PAG样和PV样免疫反应阳性神经元 ,吻尾方向几乎在其全长出现 ,多为大的假单极神经元 (直径为 2 5~ 5 0 μm)。大约 90 %左右的PAG样阳性神经元同时呈GABAARα3样和PV样免疫阳性。在激光共聚焦显微镜下观察到 ,密集分布的GAD样阳性终扣聚集在PAG样和GABAARα3样双重标记的Vme神经元胞体周围 ,并与之形成密切接触。 结论 GA BA能神经元投射至Vme的神经终末可能通过位于Vme神经元胞体内的GABAARα3亚单位对谷氨酸介导的面口部本体感觉信息的传递发挥抑制性调控作用
Objective To observe the expressions of glutamate decarboxylase (GAD) -like terminals, phosphoglycemic glutaminase (PAG), GABAA receptor α3 subunit (GABAARα3) and small albumin in rat middle trigeminal nucleus (Vme) (PV) -like positive neurons. Methods Immunofluorescence histochemical triple staining technique was observed under confocal laser scanning microscope. As a result, a large number of GABAARα3-like, PAG-like and PV-like immunoreactive neurons were observed in Vme. The kurtosis occurred nearly in its full length, mostly in the form of large pseudomorphic neurons (25-25 mm in diameter μm). About 90% of PAG-like neurons are both GABAARα3-like and PV-like immunopositive. Confocal laser scanning microscopy revealed that densely distributed GAD-like positive terminators were clustered around and in close contact with PAG-like and GABAARα3-like dual-labeled Vme neuronal soma. CONCLUSIONS: GABAARα3 subunit, located in the cell body of Vme neurons, may exert an inhibitory regulatory effect on glutamate-mediated sensory information of facial or parietal nociception in GA terminal neurons projecting to Vme
