目的探讨绿原酸对光老化人皮肤成纤维细胞ESF-1光损伤的保护作用,及其对细胞外调节蛋白激酶(ERK)信号通路的影响。方法用20 J/cm2的长波紫外线(UVA)照射ESF-1细胞制备光老化模型,以不同浓度绿原酸处理光老化细胞,检测细胞中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性、丙二醛(MDA)量,RT-PCR法检测细胞中ERK1、ERK2、基质金属蛋白酶1(MMP-1)及基质金属蛋白酶抑制因子1(TIMP-1)mRNA的表达量,Western blotting法检测p-ERK1/2蛋白的表达,酶联免疫法检测细胞上清液中MMP-1、TIMP-1的量。结果 UVA照射ESF-1细胞后,SOD活性、GSH-Px活性、TIMP-1 mRNA的表达量、TIMP-1的量明显降低,MDA水平、ERK1、ERK2、MMP-1 mRNA的表达量、p-ERK1/2、MMP-1水平明显升高(P<0.01);10.0、1.0μmol/L绿原酸能够显著改善光老化ESF-1细胞的损伤(P<0.05、0.01)。结论绿原酸调控光老化ESF-1细胞ERK信号通路,从而减轻UVA对ESF-1细胞的损伤。
Objective To investigate the protective effect of chlorogenic acid on the photodamage of ESF-1 cells and its effect on the extracellular regulated protein kinase (ERK) signaling pathway in photoaging human dermal fibroblasts. Methods The photo-aging model was established by irradiating ESF-1 cells with UVA at 20 J / cm2. The photo-aging cells were treated with different concentrations of chlorogenic acid, and the activities of superoxide dismutase (SOD), glutathione peroxidase The activity of GSH-Px, the content of malondialdehyde (MDA) and the expression of ERK1, ERK2, MMP-1 and TIMP-1 mRNA The expression of p-ERK1 / 2 protein was detected by Western blotting. Results After UVA irradiation, the activity of SOD, the activity of GSH-Px, the expression of TIMP-1 mRNA, the amount of TIMP-1 and the expression of ERK1, ERK2 and MMP-1 mRNA in ESF- ERK1 / 2 and MMP-1 levels were significantly increased (P <0.01). 10.0, 1.0μmol / L chlorogenic acid could significantly improve the injury of photo-aged ESF-1 cells (P <0.05,0.01). Conclusion Chlorogenic acid regulates the ERK signaling pathway in photogenerated ESF-1 cells and thus reduces the damage of UVA to ESF-1 cells.
