目的:研究瓣蕊唐松草根茎总生物碱对体外培养的肿瘤细胞株生长与集落形成的抑制影响。方法:用90%乙醇提取,D106大孔吸附树脂分离,乙醇洗脱得到瓣蕊唐松草根茎总生物碱。采用MTT法和集落形成法检测总生物碱对人肺腺癌细胞NCI-446、人大肠癌细胞SWWC116和人宫颈癌细胞Hela的生长与集落形成的抑制情况。结果:瓣蕊唐松草根茎总生物碱对NCI-446和SWWC116的生长与集落形成有明显的抑制作用,生长抑制的IC50值分别为1.0mg/L和1.4mg/L,集落形成抑制的IC50值分别为0.7mg/L和1.0mg/L。而对人宫颈癌细胞Hela的生长与集落形成的抑制不明显。结论:瓣蕊唐松草根茎总生物碱在体外具有明显的抗肿瘤作用。
OBJECTIVE: To study the inhibitory effect of total alkaloids from Thalictrum trifoliate rhizome on the growth and colony formation of cultured tumor cell lines in vitro. Methods: 90% ethanol extraction, D106 macroporous adsorption resin separation, elution of ethanol to obtain Prunus trilobata rhizome total alkaloids. The inhibition of growth and colony formation of human lung adenocarcinoma cell line NCI-446, human colorectal cancer cell line SWWC116 and human cervical cancer cell line Hela by total alkaloids was detected by MTT assay and colony formation assay. Results: The total alkaloids of Thalictrum palustris showed a significant inhibitory effect on the growth and colony formation of NCI-446 and SWWC116. The IC50 values of growth inhibition were 1.0 mg / L and 1.4 mg / L, respectively. The IC50 value of inhibition of colony formation Respectively 0.7mg / L and 1.0mg / L. However, the inhibition on the growth and colony formation of human cervical cancer cell Hela was not obvious. Conclusion: The total alkaloids of Tricholoma matsutake rhizome has obvious anti-tumor effect in vitro.
